مطالعه فیلوژنی مولکولی سویه سمی Anabaena sp. B3 جدا شده از دریاچه لواسان (تهران)

چکیده

دریاچه لواسان واقع در شمال‌شرق تهران، اکوسیستم پیچیده‌ای متشکل از سویه‌های سیانوباکتری‌ها با توانایی تولید طیف وسیعی از ترکیبات طبیعی سمی است که ممکن است منجر به بروز مشکلات فراوان و یا حتی مرگ انسان‌ها و حیوانات شوند. با توجه به گزارش حضور سویه‌های سمی سیانوباکتریایی در دریاچه لواسان، در این مطالعه، بررسی‌های ریخت‌شناختی و فیلوژنی سویه سمی خالص‌سازی شده از آب دریاچه مذکور، با استفاده از نشانگرهای ژنی 16S rRNA و آی.تی.اس. صورت گرفت. براساس نتایج به‌ دست آمده از درخت فیلوژنی رسم شده مبنی بر ژن 16S rRNA، سویه‌های غیرسمی با سویه سمی Anabaena sp. B3 (CCC B3)، درون یک کلاد خوشه‌بندی نشدند. به علاوه، به نظر می‌رسد که توانایی تولید سم در طول زمان از بین رفته است. نتایج حاصل از آنالیز ژن آی.تی.اس. با استفاده از Mfold نشان داد که مهم‌ترین تفاوت سویه سمی Anabaena sp. B3 با سایر سویه‌های غیرسمی Anabaena sp.، تعداد نوکلئوتیدهای لوپ انتهایی دو طرفه و تعداد لوپ‌های دو طرفه میانی در مارپیچ D1-D1 است. این در حالی است که ساختمان مارپیچ B Box در هیچ‌کدام از سویه‌ها تفاوتی نداشت. مطالعه حاضر، جزو نخستین تحقیقات انجام شده روی فیلوژنی مولکولی سیانوباکتریوم سمی در دریاچه لواسان است.
