شناسایی مولکولی مخمرهای بازیدیومیستی در خاک‌های ایران

چکیده

در این مطالعه، تنوع زیستی مخمرهای بازیدیومیستی خاک­های نواحی مختلف ایران مورد برسی قرار گرفت. برای این منظور، نمونه‌های خاک از عمق 15-5 سانتی­متری جمع‌آوری و متعاقبا مخمرها جداسازی و خالص­سازی شدند. گروه­بندی اولیه جدایه­های مخمر از طریق انگشت‌نگاریDNA  با استفاده از MSP-PCR صورت گرفت. یک یا چند جدایه منتخب از هر گروه با استفاده از داده­های توالی نواحی D1/D2، ژن S rRNA26 و تست­های فیزیولوژیک شناسایی شدند. نتایج این مطالعه نشان داد که تنوع قابل توجهی در مخمرهای بازیدیومیستی خاک‌های مناطق نمونه‌برداری شده وجود دارد. در این مقاله، تعداد 25 گونه زیر متعلق به شش جنس نخستین بار برای مایکوبیوتای ایران معرفی می‌شوند: Trichosporon pullulans، Rhodotorul mucilaginosa، R. minuta، R. lysiniphila،R. laryngis، Rhodosporidium kratochvilovae، Rh. babjevae، Holtermanniella takashimae،Fibulobasidium inconspicuum  Cryptococcus ramirezgomezianus، C. watticus، C. victoriae، C. uzbekistanensis، C. terreus، C. saitoi، C. podzolicus،C. oeirensis، C. friedmannii، C. flavescens، C. diffluens، C. bhutanensis، C. albidus، C. albidosimilis،  aerius.C، adeliensis .C. در بین جنس­های شناسایی شده، Cryptococcusبیشترین فراوانی را به خود اختصاص داد وC. aerius به عنوان گونه غالب از تمام خاک­های نمونه­برداری شده جداسازی شد.
