Two endophyte containing tall fescue genotypes (75 and 83) and their endophyte free ramets were used in this study. Endophytes were isolated from tall fescue genotypes 75 and 83 and used in three methods of inoculation including Latch and Christensen approach, insertion of mycelium into the seeds, and Johonson-Cicales approach. Endophytes were detected in new host using microscopic examination of stained tissues, AFLP marker and Koch’s postulates. Results showed that Latch and Christensen approach was the only successful technique by which cross inoculation in the two tall fescue genotypes was feasible. Success in inoculation seems to be highly dependent on splitting procedure and seedlings age. Detection methods used to confirm the presence of inoculated endophyte in plants indicated that microscopic examination is highly accurate in detecting endophytes in new host.  رابطه همزیستی بین گراس‌ها و قارچ‌های اندوفیت یکی از مهمترین همزیستی‌ها در طبیعت می‌‌باشد. قارچ‌های جنس <em>Neotyphodium</em> عضو تیره Clavicipitaceae هستند که با گراس‌های زیرتیره Pooideae همزیست می‌باشند. در این پژوهش، دو ژنوتیپ گیاهی انتخاب و قارچ‌های اندوفیت همزیست از آن‌ها استخراج گردید. وجود قارچ‌های اندوفیت در میزبان‌های تلقیح شده با روش‌های میکروسکوپی، نشانگر مولکولی AFLP و  اجرای اصول کخ بررسی شدند. در این تحقیق روش گزارش شده <em>لچ</em> و <em>کریستنسن</em> با موفقیت همراه بود و تنها از طریق این روش، قارچ‌های اندوفیت گرفته شده از یک ژنوتیپ با موفقیت وارد همان ژنوتیپ و دیگر ژنوتیپ‌ها شدند. این بررسی مشخص نمود برای تشخیص وجود قارچ در میزبان‌های گیاهی مایه‌زنی شده, روش‌های میکروسکوپی مناسب می‌باشد. https://rostaniha.areeo.ac.ir/article_101726_1f42de39f16b8322f9a534fbe137761b.pdf
